Regeneron strikes US$2.32B partnership with Parabilis to advance next-generation peptide therapies

Under the collaboration, the companies will evaluate Helicons both as standalone therapies and as payloads linked to Regeneron antibodies.

USA—Regeneron Pharmaceuticals has entered a major research collaboration with Parabilis Medicines valued at up to US$2.32 billion to develop a new generation of targeted therapies using Parabilis’ Helicon peptide platform.

The agreement will focus on creating therapeutic candidates known as antibody-Helicon conjugates (AHCs), a novel class of medicines designed to reach difficult intracellular disease targets.

The partnership reflects growing industry interest in therapies capable of targeting proteins that traditional drugs have struggled to address.

Unlike conventional small molecules, Helicons are stabilised alpha-helical peptides engineered to penetrate cells and bind to intracellular proteins, including those with broad and flat surfaces that are often considered “undruggable.”

Expanding Regeneron’s Drug Discovery Capabilities

Under the collaboration, the companies will evaluate Helicons both as standalone therapies and as payloads linked to Regeneron antibodies.

By combining antibody precision with Helicon technology, the companies aim to selectively deliver therapeutic payloads into targeted cells while modulating disease-causing proteins inside those cells.

George Yancopoulos, board co-chair, president and chief scientific officer at Regeneron, said the collaboration aligns with the company’s strategy of investing in diverse scientific platforms to broaden its pipeline of innovative medicines.

He noted that Helicons could help address previously inaccessible drug targets while also opening opportunities for entirely new therapeutic approaches.

According to Yancopoulos, coupling Helicons with Regeneron’s VelocImmune-derived antibodies may create a new therapeutic class with applications across multiple disease areas.

The collaboration will involve joint discovery efforts to develop new Helicons and AHCs, while Regeneron will oversee clinical development, manufacturing, and global commercialisation activities.

Financial terms and strategic growth

As part of the agreement, Parabilis could receive up to US$125 million in near-term funding, including a US$50 million upfront payment and a US$75 million investment tied to the company’s upcoming equity financing, subject to agreed conditions.

In addition, the deal includes up to US$2.2 billion in milestone payments as well as tiered royalties linked to future product sales.

Regeneron also retains the option to pursue additional targets through further payments.

The collaboration comes during a period of aggressive expansion for Regeneron’s research portfolio.

Just weeks earlier, the company signed another high-value agreement with Telix Pharmaceuticals, valued at up to US$4.3 billion, to develop and commercialise radiopharmaceutical therapies for cancer treatment.

Regeneron has also continued to strengthen its broader pipeline in oncology, immunology, and genetic medicine through partnerships and internal development programmes.

Recent company updates highlighted ongoing investments in advanced biologics, cell-targeting technologies, and precision medicine platforms to accelerate drug discovery and improve treatment delivery across complex diseases.
