Novartis acquires breakthrough breast cancer treatment in USD3B deal

The acquisition involves Novartis purchasing Pikavation Therapeutics, a wholly owned subsidiary of Synnovation Therapeutics that holds the rights to SNV4818 and related research programs.

USA—Swiss pharma giant Novartis has agreed to acquire SNV4818, an investigational drug designed to treat advanced breast cancer, from Synnovation Therapeutics.

The pharmaceutical giant will pay USD2 billion upfront, with an additional USD1 billion contingent on achieving specific development milestones.

Strategic acquisition details

The acquisition involves Novartis purchasing Pikavation Therapeutics, a wholly owned subsidiary of Synnovation Therapeutics that holds the rights to SNV4818 and related research programs.

This transaction represents a significant investment in oncology innovation and expands Novartis’ portfolio of cancer treatments.

SNV4818 is an oral medication currently undergoing Phase I/II clinical trials for breast cancer and other advanced solid tumors.

The drug functions as a pan-mutant selective Phosphoinositide 3-kinase alpha (PI3Kα) inhibitor, targeting a key pathway involved in tumor growth and survival.

Addressing unmet needs in breast cancer

Novartis plans to develop SNV4818 specifically for patients with hormone receptor-positive (HR+), human epidermal growth factor receptor 2 negative (HER2-) breast cancer.

This particular form of breast cancer represents a substantial patient population requiring more effective treatment options.

The company intends to explore SNV4818 as part of combination therapy strategies alongside existing treatments.

These include cyclin-dependent kinase (CDK) inhibitors and endocrine therapies, which are already used in breast cancer management.

Mutated PI3Kα serves as a well-established driver in HR+/HER2⁻ breast cancer.

However, physicians have faced challenges in effectively blocking this pathway while maintaining tolerable side effects for patients.

Innovative therapeutic approach

Shreeram Aradhye, development president at Novartis, explained that SNV4818 employs novel mutant-selective chemistry to target tumor cells more precisely.

This selectivity allows the drug to attack cancer cells while minimizing damage to healthy tissue.

Preclinical studies have shown strong activity against common mutations in the Phosphatidylinositol-4,5-bisphosphate 3-kinase catalytic subunit alpha (PIK3CA).

These genetic alterations frequently occur in breast cancer and contribute to disease progression.

Aradhye emphasized that this precision medicine approach has the potential to improve tolerability and provide more durable benefits for patients.

By targeting the underlying tumor biology more accurately, the treatment may offer better outcomes with fewer adverse effects.

Timeline and regulatory progress

Novartis expects to finalize the acquisition during the first half of 2026, pending standard closing conditions and regulatory approvals.

Clinical evaluation of SNV4818 continues to advance through ongoing trials.

Recent FDA approval

In related company news, Novartis recently received US Food and Drug Administration (FDA) approval for Cosentyx (secukinumab).

The agency authorized the medication to treat pediatric patients aged 12 years and older with moderate to severe hidradenitis suppurativa, a chronic skin condition.

This approval demonstrates Novartis’ continued success in bringing new treatments to underserved patient populations across multiple therapeutic areas.
