Regeneron expands oncology partnership with CytomX to US$4B for next-generation cancer biologics

USA—Regeneron Pharmaceuticals and CytomX Therapeutics have broadened their research and licensing partnership to accelerate the development of conditionally activated bispecific antibody therapies for cancer.

The expanded agreement increases the deal’s total potential value to up to US$4 billion and deepens their joint focus on tackling hard-to-treat tumour types that have shown limited response to conventional immunotherapy.

Established in 2022, the collaboration combines CytomX’s Probody masking technology with Regeneron’s Veloci-Bi bispecific antibody platform.

By combining these complementary approaches, the companies aim to design therapies that remain inactive in healthy tissues and become activated only within the tumour microenvironment.

Tumour-activated therapeutic design

The next-generation therapies under development are engineered to stay “switched off” until they encounter specific proteases present in tumour tissue.

Once activated, they are intended to direct immune cells precisely toward cancer cells, enhancing tumour killing while reducing damage to healthy tissues.

This strategy is particularly relevant for tumour types that have proven resistant to existing T-cell engager therapies and other immuno-oncology approaches.

By improving targeting precision, the companies hope to expand the therapeutic reach of bispecific antibodies while improving safety profiles.

Financial structure and development roles

Under the revised agreement, Regeneron will take the lead in funding preclinical, clinical, and potential commercial development activities.

CytomX will receive a US$37 million target nomination payment for two newly selected targets.

In addition, Regeneron retains the option to select up to six additional targets over the course of the collaboration.

CytomX will remain eligible for milestone payments tied to development progress, as well as for global net sales royalties, if the therapies reach the market.

The structure reflects a shared risk-and-reward model designed to support long-term oncology innovation.

Leadership perspectives on the partnership

Regeneron’s Senior Vice President of Oncology and Antibody Research, John Lin, emphasized that cancer continues to require more precise and adaptable treatment strategies.

He noted that the expanded collaboration with CytomX allows both companies to advance bispecific therapies in areas where they see the strongest scientific and clinical potential.

He also highlighted the role of Regeneron’s proprietary VelociSuite technologies in accelerating drug discovery and development efforts.

CytomX Chairman and Chief Executive Officer Sean McCarthy described the partnership as a continuation of a shared commitment to advancing cancer immunotherapy through cutting-edge science.

He explained that CytomX continues to refine its understanding of protease biology and masking technology to unlock new therapeutic opportunities, while also leveraging Regeneron’s expertise in bispecific antibodies to expand the PROBODY platform’s potential.

Broader oncology strategy and pipeline expansion

The expanded collaboration comes amid a broader wave of oncology-focused partnerships for Regeneron, which continues to strengthen its immuno-oncology pipeline through multiple external research alliances.

Earlier agreements in the same strategic direction have emphasized novel modalities such as bispecific antibodies and peptide-based platforms, reinforcing the company’s push toward next-generation targeted cancer therapies.
